ABSTRACT

@#Given the opposing effects of Akt and AMP-activated protein kinase (AMPK) on metabolic homeostasis, this study examined the effects of deletion of Akt2 and AMPKα2 on fat diet-induced hepatic steatosis. Akt2–Ampkα2 double knockout (DKO) mice were placed on high fat diet for 5 months. Glucose metabolism, energy homeostasis, cardiac function, lipid accumulation, and hepatic steatosis were examined. DKO mice were lean without anthropometric defects. High fat intake led to adiposity and decreased respiratory exchange ratio (RER) in wild-type (WT) mice, which were ablated in DKO but not Akt2−/− and Ampkα2−/− mice. High fat intake increased blood and hepatic triglycerides and cholesterol, promoted hepatic steatosis and injury in WT mice. These effects were eliminated in DKO but not Akt2−/− and Ampkα2−/− mice. Fat diet promoted fat accumulation, and enlarged adipocyte size, the effect was negated in DKO mice. Fat intake elevated fatty acid synthase (FAS), carbohydrate-responsive element-binding protein (CHREBP), sterol regulatory element-binding protein 1 (SREBP1), peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α), peroxisome proliferator-activated receptor-α (PPARα), PPARγ, stearoyl-CoA desaturase 1 (SCD-1), phosphoenolpyruvate carboxykinase (PEPCK), glucose 6-phosphatase (G6Pase), and diglyceride O-acyltransferase 1 (DGAT1), the effect was absent in DKO but not Akt2−/− and Ampkα2−/− mice. Fat diet dampened mitophagy, promoted inflammation and phosphorylation of forkhead box protein O1 (FoxO1) and AMPKα1 (Ser485), the effects were eradicated by DKO. Deletion of Parkin effectively nullified DKO-induced metabolic benefits against high fat intake. Liver samples from obese humans displayed lowered microtubule-associated proteins 1A/1B light chain 3B (LC3B), Pink1, Parkin, as well as enhanced phosphorylation of Akt, AMPK (Ser485), and FoxO1, which were consolidated by RNA sequencing (RNAseq) and mass spectrometry analyses from rodent and human livers. These data suggest that concurrent deletion of Akt2 and AMPKα2 offers resilience to fat diet-induced obesity and hepatic steatosis, possibly through preservation of Parkin-mediated mitophagy and lipid metabolism.

ABSTRACT

In view of the pain points and difficulties in the current stroke service system, the Internet-plus model is merged into the stroke service process for exploration and practice. Mobile Internet technology bridged the process before and after the patient arrives at the hospital, thus extending the green path. This green path is under refined management by mobile applications and 2-dimensional bar code technologies, supporting sustained improvement of data-driven clinical quality. Benefited by"Internet-plus"stroke service management, the efficiency and key performance indicator of the stroke center are significantly elevated.

ABSTRACT

@#BACKGROUND: Noninvasive monitoring of intra-abdominal pressure (IAP) by measuring abdominal wall tension (AWT) was effective and feasible in previous postmortem and animal studies. This study aimed to investigate the feasibility of the AWT method for noninvasively monitoring IAP in the intensive care unit (ICU). METHODS: In this prospective study, we observed patients with detained urethral catheters in the ICU of Shanghai Tenth People's Hospital between April 2011 and March 2013. The correlation between AWT and urinary bladder pressure (UBP) was analyzed by linear regression analysis. The effects of respiratory and body position on AWT were evaluated using the paired samples t test, whereas the effects of gender and body mass index (BMI) on baseline AWT (IAP<12 mmHg) were assessed using one-way analysis of variance. RESULTS: A total of 51 patients were studied. A significant linear correlation was observed between AWT and UBP (R=0.986, P<0.01); the regression equation was Y=–1.369+9.57X (P<0.01). There were significant differences among the different respiratory phases and body positions (P<0.01). However, gender and BMI had no significant effects on baseline AWT (P=0.457 and 0.313, respectively). CONCLUSIONS: There was a significant linear correlation between AWT and UBP and respiratory phase, whereas body position had significant effects on AWT but gender and BMI did not. Therefore, AWT could serve as a simple, rapid, accurate, and important method to monitor IAP in critical y il patients.

ABSTRACT

K-complex (KC) is a major component of electroencephalography (EEG) in human brain during non-rapid eye movement sleep, and therehas long been a debate on its function. Some researchers consider that KC represents a sign of arousal response of brain to internal or external stimuli, and others suggest that it is a protective response to sustain sleep. In this article we put forward a functional model of KC based on the above two views and related literatures. According to our model, KC arises from three kinds of brain rhythms (sleep cyclic, central oscillation, and cortical oscillation). Based on the results of stimulus significance assessed by sleep-information assessment center, the generation of KC is related to three different responses-total arousal, sign of being awakened, and no sign of being awakened. In conclusion, KC is a reflection of cerebral state responsive to stimuli.

ABSTRACT

Objective To detail the utility of CT scan in detection of urinary stones induced by melamine tainted formula. Material and Methods A total of 1062 children fed with melamine-contaminated infant formula were screened for urinary stones in our institute from September through December 2008. Ultrasonography of the urinary tract system was performed in all these children. If the children with suspected stones or severe obstruction were presented after ultrasound examination, themulti-detector row CTurographic examination was advocated subsequently. Results Ultrasound examination in combination with multidetector row CT urography could increase the diagnostic rate from 3.4% (36/1062) by ultrasound examination alone to 4.6% (49/1062). Conclusions The specificity and sensitivity of the multidetector row CT urographic examination is higher than ultrasonography.

ABSTRACT

The high frame rate (HFR) ultrasonic imaging system based on linear frequency-modulated (LFM) signal constructs images at a high frame rate; the signal-to-noise ratio (SNR) of this system can also be improved. Unfortunately, such pulse compression methods that increase the SNR usually cause range sidelobe artifacts. In an imaging situation, the effects of the sidelobes extending on either side of the compressed pulse will be self-noise along the axial direction and masking of weaker echoes. The improvement on high frame rate ultrasonic imaging system based on LFM signal is considered in this paper. In this proposed scheme, a predistorted LFM signal is used as excited signal and a mismatched filter is applied on receiving end. The results show that the proposed HFR ultrasonic imaging system can achieve higher SNR and the axial resolution is also improved.

ABSTRACT

The high frame rate (HFR) ultrasonic imaging system, which is developed with limited diffraction beams, constructs images at a high frame rate. However the rectangular imaging area, to some extent, restricts the far field imaging information. At the same time, because of one transmission for constructing image, the system suffers from low SNR. In this paper we present a computationally efficient method to construct sector mode image and to increase the SNR in HFR system. The method uses Golay complementary sequence as excitation to realize two transmission events. Each event simultaneously transmits two plane waves with different transmission angle. Then the received echo signals related to different angle are separated according to orthogonality of Golay complementary sequence and used to construct two images of different area by HFR method. Finally the two images are synthesized to one frame of sector mode image.

ABSTRACT

In order to study event-related desynchronization (ERD) related to voluntary movement, we designed two experiments. In the first experiment, untrained subjects were required to imagine the action of typing with left or right index finger for about 1 second before real action, whereas they were required to type instantly after instruction in the second experiment. By analyzing spontaneous EEG signals between the instruction and the action, we predicted which finger was used. The prediction accuracy in the first experiment fell from 85% to 71% with the progress of experiment, the average accuracy being 78%, whereas the prediction result was almost random guess in the second experiment. The results demonstrate that (1) ERD patterns are significantly affected by the effective duration of motion imagination, (2) unconscious reduction of this duration can decrease the prediction accuracy. Therefore, when designing subsequent BCI experiments, we should devote our attention to the question of how to keep the effective duration of motion imagination.

ABSTRACT

A high frame rate (HFR) ultrasonic imaging method developed by Jianyu Lu has been attracting attention in the related imaging area. Because a special weighting on the received signal is needed to implement this method, the imaging system is complicated. This paper presents a new kind of theroetical analysis for the HFR imaging system in view of the angular spectrum concept. Based on the study, the special weighting process can be replaced by Fourier transform. Computer simulation demonstrates the correctness of the analysis. It shows that the constructed images by the new method have the same high quality as those obtained by using the original HFR method while the system implementation is greatly simplified.

ABSTRACT

The simulation of excitation propagation's process in human heart is one of the main aspects of ECG forward problem. The simulation results not only are the criterion of the simulation model's precision and reliability, but also have great value in researches and diagnoses. We performed the simulation of QRST waves of complete left bundle branch block (LBBB) and right bundle branch block (RBBB) in virtue of a vector propagation algorithm (VPA), which is accurate, efficient and applicable to anisotropic computer heart models. The simulation results accord with the actual QRST wave in clinical practice.
